Mammalian expression vectors

 

pDSG-IBA vectors

pDSG-IBA is a small transient expression vector especially developed for the use in combination with the MEXi mammalian expression system. Efficient extrachromosomal replication due to EBV oriP driven by EBNA-1. Requires MEXi 293E cell line or any other cell line expressing EBNA-1 gene.

  • CMV promoter: high level constitutive expression

  • oriP: origin of replication from Epstein-Barr Virus for extrachromosomal replication driven by EBNA-1 (MEXi 293E cells express EBNA-1)

  • ColE1 + Amp: for propagation in E. coli

  • BM40: secretion signal to transfer the expressed protein into the medium (not on all vectors available)

 

pESG-IBA vectors

pESG-IBA is a small expression vector recommended for transient expression of proteins in cell lines providing SV40 large T-antigen for extrachromosomal replication with SV40 ori. NeoR for generation of stable cells lines.

  • CMV promoter: high level constitutive expression

  • SV40 ori: origin of replication for extrachromosomal replication driven by SV40 large T-antigen (as provided by HEK293T or COS-1, -3, -7 cells)

  • ColE1 + Amp: for propagation in E. coli

  • NeoR: neomycin resistance gene to generate stable cell lines

  • BM40: secretion signal to transfer the expressed protein into the medium (not for all vectors available)

pCSG-IBA vectors

pCSG-IBA is a large expression vector with universal features for transient expression as well as for generation of stable cell lines (NeoR). Extrachromosomal replication by either the more powerful oriP (cell line independent due to EBNA-1 gene provided by vector ) or SV40 ori (cell line dependent) possible.

  • CMV promoter: high level constitutive expression

  • oriP: origin of replication from Epstein Barr Virus for extrachromosomal replication driven by EBNA-1

  • EBNA-1 gene: Expression of EBNA-1 for initiation of extrachromosomal replication by oriP

  • SV40 ori: origin of replication for extrachromosomal replication driven by SV40 large T-antigen (as provided by HEK293T or COS-1, -3, -7 cells)

  • NeoR: neomycin resistance gene to generate stable cell lines

  • ColE1 + Amp: for propagation in E. coli
