So far, Strep-Tactin® Superflow® high capacity is one of the most favored resins allowing the purification of recombinant proteins in high yields. However, the new Strep-Tactin®XT Superflow® high capacity provides significant more purified protein because it combines an increased capacity of the resin with a very efficient binding due to the high affinity for Strep-tag®II and Twin-Strep-tag®.

Four different proteins fused to either Twin-Strep-tag® or Strep-tag®II were purified using Strep-Tactin® Superflow® high capacity and Strep-Tactin®XT Superflow® high capacity. Each lysate was applied  as concentrated extract (3 mg/ml) and as diluted cell extract (0.4 mg/ml), respectively. Strep-Tactin®XT Superflow® high capacity has the highest binding capacity in both experiments. The observed binding capacities for Strep-Tactin®XT Superflow® high capacity were 1.5 – 3.4 fold higher in experiments using diluted cell extractscompared to Strep-Tactin® Superflow® high capacity, and on average 1.8 fold high if using concentrated extracts. Remarkably, lower protein concentrations in the feedstock have no significant influence on the binding capacity of Strep-Tactin®XT Superflow® high capacity, independent of the used tag. In contrast, a significant decreased binding capacity is observed for Strep-Tactin® Superflow® high capacity at the lower cell extract concentration.
