Specifications for Strep-Tactin® Purification Resins

There are several Strep-Tactin® resin versions available. All resins differ in their properties and suitability for applications. Learn more about the different resin matrices in order to know when to use which resin.

Features The Strep-Tactin® Superflow high capacity binds in comparison to Strep-Tactin® Superflow a three to five fold higher amount of target protein and thereby achieves a higher yield. Strep-Tactin® Sepharose is a good alternative for large proteins, if the affinity of Strep-Tactin®XT 4Flow® is too high.  The alternative when cell components (e.g. chlorophyll from plant extracts) bind non-specifically to Sepharose, Superflow or 4Flow®. Strep-Tactin®XT Superflow high capacity achieves higher yields than Strep-Tactin® Superflow high capacity and thereby is more cost efficient. Strep-Tactin®XT 4Flow® is the resin to start with. Due to the higher affinity compared to Strep-Tactin® resins it is particularly applicable for low abundant or challenging proteins.
If the affinity of Strep-Tactin®XT resins is too high and the target protein is not fully eluted, then Strep-Tactin® Superflow is a good alternative. Only suitable for gravity flow chromatography due to the low pressure stability. Synthetic support in comparison to all other resins. Can be used for protein purification under denaturing  conditions (6 M urea). Suitable for all types of proteins and especially for large proteins, because it has the high affinity of Strep-Tactin®XT, the broadest pH stability and consists of a low-concentrated agarose.

** Dynamic binding capacity (DBC) was determined with mCherry-Twin-Strep-tag® (30 kDa). The DBC is determined under realistic operation conditions and shows the amount of protein which is bound until 10% of the protein is found in the flow through.

Strep-Tactin® Sepharose®

For purification of Strep-tag®II proteins by gravity flow column chromatography

Strep-Tactin® Sepharose® provides high binding capacity and minimal non-specific binding.
The material can be used for gravity flow purification. Since the Strep-tag® protein purification should be performed as column affinity chromatography, we recommend our pre-packed Strep-Tactin® Sepharose® gravity flow columns, which are available in different formats (from 0.2 ml mini columns up to 10 ml columns).

For packing individual columns bulk material is available.

For batch purification we recommend to use magnetic beads: MagStrep "type3" XT beads.

Strep-Tactin® Superflow®

Strep-Tactin® Superflow® resin:

  • combines outstanding flow characteristics with superior mechanical stability
  • is suitable for increased flow rates and useful for purification of large protein complexes
  • yields much higher protein purity (> 95%) than 6xHistidine-tag purification on Ni-NTA Superflow®

Strep-Tactin® Superflow® high capacity resin offers a three times higher binding capacity at identical high protein purity. It is the solution for Strep-tag® fusion proteins at high expression levels.

NEW: Strep-Tactin®XT Superflow® high capacity

So far, Strep-Tactin® Superflow® high capacity is one of the most favored resins allowing the purification of recombinant proteins in high yields. However, the new Strep-Tactin®XT Superflow® high capacity provides significant more purified protein because it combines an increased capacity of the resin with a very efficient binding due to the high affinity for Strep-tag®II and Twin-Strep-tag®.

Strep-Tactin® MacroPrep®

The MacroPrep® resin exhibits non-specific binding properties differing from those of Sepharose® or Superflow® and can, thus, be recommended in cases where suboptimal results are obtained with Sepharose® or Superflow®, and vice versa.

MacroPrep® is available in one format:

  • 50% suspension (slurry)
